Sanger Sequencing, also known as the chain-termination method or dideoxy sequencing, is a classic and highly reliable molecular technique used for precise determination of the nucleotide sequence of a specific DNA segment. It involves replicating a DNA template strand using DNA polymerase, a primer, deoxynucleotides (dNTPs), and a small proportion of dideoxynucleotides (ddNTPs). The ddNTPs lack a 3'-hydroxyl group, preventing further elongation of the DNA strand once incorporated, thus acting as chain terminators. This process generates a series of DNA fragments of varying lengths, each terminated at a specific nucleotide. These fragments are then separated by size (traditionally by gel electrophoresis or, more commonly now, by capillary electrophoresis) and detected by fluorescent labels, revealing the DNA sequence. Sanger sequencing is considered the "gold standard" for confirming variants detected by other methods, for targeted analysis of specific genes or exons where a mutation is strongly suspected, and for identifying point mutations and small insertions/deletions. It provides high accuracy (typically >99.9%) and reads lengths up to ~1000 base pairs per reaction.
