MLPA (Multiplex Ligation-dependent Probe Amplification)
MLPA (Multiplex Ligation-dependent Probe Amplification) is a highly sensitive and robust molecular technique used for the detection of copy number variations (CNVs), which include gene deletions or duplications. It allows for the simultaneous detection of up to 50 different genomic sequences in a single reaction. The principle relies on the hybridization of specifically designed probes to target DNA sequences. Each probe consists of two oligonucleotides that, when bound adjacently to the target sequence, can be ligated. The ligated probes are then amplified by PCR using universal primers. The amount of each amplified product is directly proportional to the number of target DNA copies present in the sample. By comparing the peak heights or areas of the amplified fragments from a test sample to those from a normal reference sample, gains or losses of specific genomic regions (exons, entire genes, or chromosomal segments) can be accurately identified. MLPA is particularly valuable for detecting conditions caused by single or multiple exon deletions/duplications that are often missed by traditional Sanger sequencing or standard NGS panels which primarily focus on point mutations and small indels.
