Quantitative RT-PCR (RT-qPCR), also known as real-time RT-PCR, is a highly sensitive and precise molecular diagnostic technique used to quantify the amount of a specific RNA sequence. For the BCR/ABL (CML) Major (p210) test, RT-qPCR first converts the messenger RNA (mRNA) of the fusion gene into complementary DNA (cDNA) using reverse transcriptase. This cDNA is then amplified, and the accumulation of the amplified product is measured in real-time using fluorescent probes. This allows for the precise and reproducible quantification of the BCR/ABL fusion gene transcripts relative to a control gene (e.g., ABL or GUS), providing an accurate measure of disease burden. This quantitative aspect is critical for monitoring treatment response and minimal residual disease (MRD) in CML patients.
